Candidate Genes
Microarray gene expression analysis was used to identify candidate genes for QTLs associated with collagen-induced arthritis. RNA from inflamed paws of collagen-treated mice were used in the microarray analysis and compared to RNA from normal paws. 223 genes showed a four-fold or greater expression difference between inflamed and normal paws. Nine of these genes are located near previously identified arthritis severity/susceptibility QTLs and may be considered for candidates. Mapping and Phenotype information for this QTL, its variants and associated markersJ:53356The authors performed experiments to localize susceptibility loci for collagen induced arthritis (CIA). F2 crosses between the highly susceptible DBA/1J strain and the highly resistant SWR/J strains were analyzed for the presence of QTLs. A locus, Cia3 on mouse Chromosome 6 had significant evidence of linkage to D6Mit10 (lod score of 3.19). Two-point analysis using BYMARKER showed that the incidence and severity of the CIA phenotype was significantly associated with D6Mit10 (F = 8.9, P<0.00045). Candidate genes in this interval include Tnfrsf1a, Cd4, Il5ra, Tnfrsf7, Tgfa and Hrh1. |
