Candidate Genes
The correlation between gene expression and dietary interaction was examined in a population of (BALB/cStCrlfC3H/Nctr x VY/WffC3Hf/Nctr-A(vy)/a)F1 animals. A/a and A/Avy animals were placed on a 70% (calorie restricted) or 100% (non-restricted) diet andliver mRNA levels were assessed for over 18,000 genes using DNA microarrays. Twenty-eight known genes showing statistically significant differential expression between the 70% and 100% calorie diets and A/a and A/Avy genotypes mapped near known diabesity QTLs. These genes may be considered further for candidate genes. Mapping and Phenotype information for this QTL, its variants and associated markersJ:51880192 (C57BL/6Jcl x KK.B6-Ay/Jcl)F2 intercross animals were typed for 97 polymorphic markers to identify QTLs associated with hyperglycemia and impaired glucose tolerance. Fglu, a QTL significantly associated with fasting glucose levels in a/a F2 animals, mapped to mouse chromosome 6 at approximately 16 cM with a QTL range from 0.68 - 29 cM and LOD = 6 at D6Mit268. Candidate genes in the region of Fglu are the leptin gene (Lep) and neuropeptide Y (Npy). Suggestive QTLs for fasting glucose levels in a/a F2 animals mapped to mouse chromosomes 3 (LOD = 2.8 at D3Mit102), 5 (LOD = 2.8 at D5Mit113), and 14 (LOD = 3.0 at D14Mit165). A suggestive QTL for fasting glucose in Ay/a F2 animals mapped to mouse chromosome 9 at D9Mit229 with a LOD score of 2.8. Suggestive QTLs for glucose tolerance in Ay/a F2 animals mapped to mouse chromosomes 1 (LOD = 2.8 at D1Mit303) and 8 (LOD = 3.0 at D8Mit191). |
