The correlation between gene expression and dietary interaction was examined in a population of (BALB/cStCrlfC3H/Nctr x VY/WffC3Hf/Nctr-A(vy)/a)F1 animals. A/a and A/Avy animals were placed on a 70% (calorie restricted) or 100% (non-restricted) diet andliver mRNA levels were assessed for over 18,000 genes using DNA microarrays. Twenty-eight known genes showing statistically significant differential expression between the 70% and 100% calorie diets and A/a and A/Avy genotypes mapped near known diabesity QTLs. These genes may be considered further for candidate genes.

On mouse Chromosome 8, Es22 (43.2 cM) and Cdh3 (53.3 cM) both show differential expression between restricted and unrestricted diets on the A/a background. Diabesity QTL Wg3 (45 cM) mapsnear Es22 and diabesity QTL Bwq3 (56 cM) maps near Chd3.

Mapping and Phenotype information for this QTL, its variants and associated markers

391 (SM/J x A/J)F2 animals were genotyped at 90 polymorphic loci to identify QTLs associated with body weight. Parental strain SM/J weighs approximately 65% less than parental strain A/J. Significant linkage to body weight was detected on mouse Chromosome 8 (Bwq3) and mouse Chromosome 18 (Bwq4). Bwq3 spans 53 cM - 69 cM on chromosome 8 with a peak LOD score of 4.6 at 56 cM near D8Mit88. Bwq3 appears to exhibit recessive or additive inheritance where SM/J-derived alleles are associated with increased bodyweight. Bwq4 spans 20 cM - 54 cM on Chromosome 18 with a peak LOD score of 4.8 at 28 cM near D18Mit15. SM/J-derived alleles appear to confer increased body weight in a dominantly inherited fashion at this locus. Early growth response 1 (Egr1) and glucocorticoid receptor 1 (Nr3c1) are possible candidate genes for Bwq4. No candidate genes have been identified in the region of Bwq3. Suggestive linkages to body weight were detected on mouse Chromosomes 2, 6, 7, 15, and 19 (LOD<3.5).
