Mapping and Phenotype information for this QTL, its variants and associated markersJ:68417The authors use a novel approach in mapping QTLs involved in murine behavioral phenotypes. A genome wide scan was performed with (C57BL/6J x DBA/2J)F2 and (BALB/cJ x LP/J)F2 intercrosses crosses to map QTLs affecting in three different behavioral phenotypes. The strains chosen have widely differing polymorphisms and phenotypes, and the QTLs identified from each cross do not appear to have overlap. Significant QTLs (LOD>4.3 threshold) for basal locomotor activity (Bslm1) and haloperidol-induced catalepsy (Hpic1) were mapped to Chromosome 9 at positions 25 - 55 cM and 20 - 50 cM, respectively. Bslm1 was identified from the (BALB/cJ x LP/J)F2 cross, and Hpic1 was identified from the (C57BL/6J x DBA/2J)F2 cross. The authors also used a subtractive method to resolve a broad QTL (60 - 100 cM) for basal activity on distal Chromosome 1. Genotyping information from (C57BL/6J x DBA/2J)F2, (C57BL/6J x A/J)F2, and (C57BL/6J x BALB/cJ)F2 intercrosses was used to screen 254 microsatellite markers across the locus. Onlymarkers polymorphic between all four strains (35 markers) were further analyzed. These highly informative markers revealed 2 separate QTLs: one previously reported by Talbot for basal activity at 67 cM and another at 95 - 105 cM. J:61419678 male animals from a (C57BL/6J x DBA/2J)F2 intercross were used to map catalepsy-related QTLs. Markers were typed at an average density of 20 cM. Hpic1, a QTL for haloperidol-induced catalepsy, mapped to 48 cM (QTL range spanning 25 cM - 55 cM) on chromosome 9 with a LOD score of 5.4 at D9Mit111. Candidate genes found at this locus include Drd2, Ncam, Acat1, Htr1b, Chrna5, Myo5a, and El1. A probable QTL for halperidol-induced catalepsy, Hpic5, mapped to 9 cM on Chromosome 1 with a LOD score exceeding the threshold of 3.5 at D9Mit188 and D9Mit90. Potential candidate genes in this region are Penk-rs and Gria4. The dilute coat color was found to segregate against the parental phenotype. Dilute DBA/2J animals have a haloperidol ED50 of 0.4 mg/kg and dilute C57BL/6J animals have a haloperidol ED50 of 4.0 mg/kg. |
