Candidate Genes

The correlation between gene expression and dietary interaction was examined in a population of (BALB/cStCrlfC3H/Nctr x VY/WffC3Hf/Nctr-A(vy)/a)F1 animals. A/a and A/Avy animals were placed on a 70% (calorie restricted) or 100% (non-restricted) diet andliver mRNA levels were assessed for over 18,000 genes using DNA microarrays. Twenty-eight known genes showing statistically significant differential expression between the 70% and 100% calorie diets and A/a and A/Avy genotypes mapped near known diabesity QTLs. These genes may be considered further for candidate genes.

On mouse Chromosome X, Atp6ap1 (29.8 cM), Flna (29.8 cM), and Gdi1 (29.8 cM) all show differential expression between A/a and A/Avy animals on the restricted diet. Diabesity QTLs Bw19 (26.4 cM) and C10bw6 (32 cM) map near these candidate genes. Jarid1c (64 cM) shows differential expression between the restricted and unrestricted diets on the A/Avy background. Diabesity QTL Dob7 (59.5 cM) maps near Jarid1c.

A total of 387 male and female mice used in a genome wide scan for Body Weight QTLs and were derived from (C57BL/6J x M.m. castaneus) F1 females backcrossed to M.m. castaneus. M.m. castaneus have body weight QTLs that are 60% lower then C57BL/6J mice at10 weeks of age.

Seven sex specific QTLs were mapped to mouse chromosomes. C10Bw1 mapped to mouse Chromosome 2 near D2Mit7 (LOD 10.7). C10Bw2 a female specific QTL mapped to chromosome 9 near D9Mit2 (LOD 3.6). C10Bw3 a male specific QTL mapped to chromosome11 near D11Mit67 (LOD 3.4). C10Bw4 a female specific QTL mapped to chromosome 13 near D13Mit26 (LOD 3.3). C10Bw5 a male specific QTL also mapped to chromosome 13 near D13Mit195 (LOD 3.7). C10Bw6 a female specific QTL mapped to the X chromosome near DXMit75 (LOD 4.3). C10Bw7 a male specific QTL mapped to Chromosome 12 near D12Mit4, but did not have a main effect and therefore was epistatic.
