Notes |
This allele is associated with higher stem cell cycling rates and shorter life span.
Mapping and Phenotype information for this QTL, its variants and associated markersJ:5411526 BXD recombinant inbred (RI) strains and 12 BXH REI strains (B = C57BL/6J, D = DBA/2J, H = C3H/HeJ) were screened for loci associated with hemopoietic S-phase cell cycling using a mapping panel. C57BL/6J progenitor strain tends to have lower cell cycling times and longer life span whereas progenitor strains DBA/2J and C3H/HeJ tend to have higher cell cycling times and shorter life spans. Scpro1 (stem cell proliferation) and Scpro2, mapped to mouse chromosome 7 at 6 cM (LOD = 3.4 at D7Ncvs38) and mouse chromosome 11 at 31 cM (LOD = 3.2 at D11Ncvs76), respectively, in the BXD RI strains and were confirmed in the BXH RI strains. DBA/2J-derived alleles at Scpro1 and Scpro2 predisposed animals to 25% shorter life spans and higher cell cycling. Scpro3, a locus associated with cell cycling mapped to mouse chromosome 10 at 60 cM (LOD = 3.7 at Iapls2-37) in the BXH RI strains and is located near the Kit1 gene. J:78926Scpro2, a QTL associated with stem cell proliferation rates, was mapped and confirmed in 33 BXD (B=C57BL/6; D=DBA/2) recombinant inbred (RI) strains and 60 C57BL/6 x (C57BL/6 x DBA/2)F1 backcross animals. Scpro2 mapped to D11Mit131 (29 CM) in the BXD RI set and to D11Mit178 (47 cM) in the backcross set. DBA/2-derived alleles are associated with higher cell proliferation rates. Scpro2 was further confirmed in congenic line B6.D2-Scpro2. This congenic carries DBA-derived DNA from 39 cM - 49 cM on Chromosome 11 on a C57BL/6 genetic background. This congenic recapitulates the high cell cycling rates of donor strain DBA/2. Potential candidate genes for Scrpo 2 are Pafah1b1 and Tbx2. |
