Mapping and Phenotype information for this QTL, its variants and associated markers

J:68419

Genome scan was performed on animals from reciprocal (NZB/BlNJ x C57BL/6By)F2 intercrosses to identify QTLs affecting sensitivity to tasting bitterness in sucrose octaacetate (SOA) solution. Parental strain NZB/BlNJ is a demi-taster whereas C57BL/6By is a nontaster. 65 microsatellite markers at an average spacing of 22.5 cM were used in the genome scan. Bitterness tasting QTLs were detected on mouse Chromosomes 2, 6, 11, and 19. Bits1 maps to mouse Chromosome 2 from 65 cM - 85 cM giving a peak LOD score of 5.8 at D2Mit42 and is associated with bitterness tasting at a low SOA concentration. The NZB/BlNJ-derived allele increases SOA consumption in a recessively inherited fashion at this locus. Possible candidate genes for Bits1 are Plcb2, Itpa, and Plcb4. Bits2 maps to mouse Chromosome 6 from 45 cM - 65 cM giving a peak LOD score of 9.4 at D6Mit9 and is associated with bitterness tasting at a high SOA concentrations. The NZB/BlNJ-derived allele increases SOA consumption in a codominant fashion at Bits2. The Bits2 locus replicates a previously identified locus for bitterness tasting, Soa. A possible candidate gene for Bits2 is Itpr1. Bits3 maps to mouse Chromosome 11 from 0 cM - 35 cM giving a peak LOD score of 6.5 at D11Mit19 and is associated with bitterness tasting athigh and low SOA concentrations. The NZB/BlNJ-derived allele increases SOA consumption in a recessively inherited fashion at this locus. Bits4 maps to mouse Chromosome 19 from 10 cM - 20 cM giving a peak LOD score of 3.2 at D19Mit16 and is associated with bitterness tasting at high and low SOA concentrations. The NZB/BlNJ-derived allele decreases SOA consumption in a dominantly inherited fashion at this locus. A possible candidate gene for Bits4 is Plcb3.
