Mapping and Phenotype information for this QTL, its variants and associated markers

J:40120

Genome scan was performed on 19 BXD (B=C57BL/6J; D=DBA/2J) recombinant inbred (RI) strains to identify QTLs associated with alcohol withdrawal severity. A locus named Alcw4 (authors refer to this locus as Aw1) contributing to alcohol withdrawal severity mapped to 37 cM on mouse Chromsome 2 in linkage to D2Mit9. This locus was then confirmed using a (C57BL/6J x DBA/2J)F2 population. DBA/2J-derived alleles confer increased alcohol withdrawal severity. F2 animals homozygous for C57BL/6J-derived alleles at D2Mit9 exhibit significantly less pronounced withdrawal symptoms that were also shorter in duration compared to F2 animals homozygous for DBA/2J-derived allele at D2Mit9. Alcw4 was also confirmed using WSP (withdrawal seizure prone) and WSR (withdrawal seizure resistant) strains. The distribution of alleles between WSP and WSR strains was disproportionate at D2Mit9 while the alleles at other loci were not. Possible candidate genes for Alcw4 include Gad1, Scn1a, Scn2a, and Scn3a.
