Mutation detailsExon 2 was replaced with a neomycin resistance gene cassette via homologous recombination. Homozygous mutant embryos were observed at a frequency of 10% and 6% at E8.5 and E9.5, respectively, by PCR genotype analysis. Because homozygous mutant animals could not be obtained, embryonic stem cell lines from E3.5 homozygous mutant embryos were established. RT-CR and Western blot analysis of the mutant stem cell lines confirmed the absence of gene expression. (J:76298)
