Mutation details: This allele was generated by breeding mice carrying Gabra1tm1Uru with transgenic mice expressing cre recombinase via the EIIa promoter. The floxed neomycin cassette was excised from intron 4, while the point mutation in codon 101 that results in a histidine to arginine substitution remained intact. Normal levels and patterns of expression were determined via immunoblotting and immunohistochemical analysis of homozygous mutant mice. Receptors encoded by the targeted locus isolated from mutant mice showed ligand-binding profiles consistant with those of diazepam-insensitive receptors.
