Mutation details: A construct containing rat Kcna1 sequence appended with a hemagglutinin tag (HA) and neo was incorporated at the enodogenous locus via homologous recombination. The rat sequence was inserted such that it would replace the Kcna5 coding sequence and be expressed by the Kcna5 promoter. Northern blot analysis confirmed the absence of Kcna5, while the presence of the rat Kcna1 was detected by RT-PCR analysis. HA epitope directed antibodies showed a 62 kDa protein in the heart tissue obtained from mutant mice, but not in brain tissue.
