Mutation detailsThe transgene comprises one to two copies of bacterial artificial chromosome (BAC) clone RP23-291P1, whose 182-kb, C57BL/6J-derived genomic DNA insert contains the Abc11 gene, 33 kb of 5' flanking and 44 kb of 3' flanking DNA, and the entire glucose-6-phosphatase, catalytic, 2 (G6pc2) gene. Northern blot analysis of total liver RNA and western blot analysis of canalicular liver plasma membrane extracts showed about two-fold elevation of Abc11 transcript and protein levels in transgenic mice fed a chow diet; transcript levels were unaffected by consumption of a lithogenic diet for 8 weeks. The primary publication does not distinguish, in describing experimental results, between this and another transgenic line, designated Tg(RP23-291P1)2Flp, generated in the same strain (129S1/SvImJ) using this BAC.

Some differences, though not statistically significant, were noted in semi-quantitative scores of gallbladder biles of transgenic vs. wild-type 129S1/SvImJ mice fed a lithogenic diet for 4 weeks for:

  • cholesterol monohydrate crystals (1.4 +/- 0.6 vs. 1.1 +/- 0.6)
  • sandy stones (0.5 +/- 0.3 vs. 0.4 +/- 0.3)
  • prevalence of true gallstones (25% vs. 14%)

Densitometric analysis of northern blots of total liver RNA from chow-fed mice revealed Cyp7a1 expression of transgenic mice to be only 35% +/- 14% that in wildtype mice.
