Mapping and Phenotype information for this QTL, its variants and associated markers

79 microsatellite markers spanning 19 chromosomes were screened in a (B10.D2 x DBA/2)F1 x DBA/2 backcross population to identify QTLs linked to graft-versus-host disease (GVHD). Lymphoid cells from backcross animals were injected into (C57BL/6 x DBA/2)F1recipients to induce GVHD. Lymphoid cells from DBA/2 donor animals induce a greater level of serum anti-ssDNA autoantibodies in recipients than lymphoid cells from B10.D2 donors. Significant linkage to anti-ssDNA autoantibody levels was detected at 40 cM on mouse Chromosome 11 near D11Mit278 (P<0.0005) and this locus was named Gvhd4. Gvhd4 also appears to be linked to the development of GVHD-associated renal pathology (P<0.002). A second locus, Gvhd5, was identified at 78.5 cM on mouse Chromosome 4 nearD4Mit226 (P<0.0005). Donors homozygous for DBA/2-derived alleles at both Gvhd4 and Gvhd5 appear to induce stronger autoantibody response in recipient animals. Donors homozygous for DBA/2-derived alleles at Gvhd4 also appear to confer increased renal pathology in recipient animals.
