12 C57BL/6J x C3H/HeJ (BXH) recombinant inbred (RI) strains were screened for 558 SSLPs to identify QTLs associated with susceptibility to ozone-induced lung hyperpermeability. Parental strain C57BL/6J is susceptible to ozone-induced injury and exhibits greater lung hyperpermeability compared to the resistant parental strain C3H/HeJ. A locus on mouse Chromosome 4, Lhyp, was identified with significant linkage to ozone-induced lung hyperpermeability (LRS=16.6) and maps to an interval between Mup1 (27.8 cM) and Tlr4 (33 cM). Suggestive loci were detected on distal chromosome 3 and central chromosome 11. Since the Tlr4 allele differs between C57BL/6J (wild type) and C3H/HeJ (mutant), this gene was investigated as a candidate for Lhyp. Substrain C3H/HeOuJ andC3H/HeJ differ only in the coding region of Tlr4. Following ozone exposure, C3H/HeOuJ (wild type Tlr4) exhibits greater lung hyperpermeability compared to C3H/HeJ (mutant Tlr4). RT-PCR analysis also revealed a difference in Tlr4 expression levels betweenthe 2 strains following ozone exposure.
