Genome scan using 93 microsatellite markers at an average resolution of 10 cM was performed on a population of 183 (C57BL/6NCrj x DBA/2NCrj)F2 animals to identify QTLs associated with experimental severe malaria (ESM) resistance/susceptibility. Parental strain C57BL/6NCrj is susceptible to ESM with a mean survival time of 6-8 days post-infection with Plasmodium bergheri and exhibits neurological symptoms such as axtia and coma. DBA/2NCrj is relatively resistant to ESM, surviving more than 20 days post-infection and exhibits no neurological pathologies. A locus on central mouse Chromosome 18, Esmr (experimental severe malaria resistance), showed significant linkage at 31 cM near D18Mit123 (P = 4.1 x 10-8). DBA/2NCrj-derived alleles confer recessively inherited resistance to EMS at this locus. Several candidate genes map to this region and include Csf1r, Pdgfrb, Cd14 and Ii. Suggestive loci mapped to mouse Chromosomes 5 (D5Mit10 and D5Mit136), 8 (D8Mit249), and 14 (D14Mit206 and D14Mit54). DBA/2NCrj-derived alleles confer resistance at the Chromosome 5 and 14 loci and susceptibility at the Chromosome 8 locus.
