Acsns2 and Acsns3 interact epistatically. Homzygosity for SM/J-derived alleles at both loci increases susceptibility to infection and decreases host survival time.
Mapping and Phenotype information for this QTL, its variants and associated markersJ:75549A whole genome scan using 95 markers at an average spacing of 20 cM was performed on 207 male (A/J x SM/J)F2 animals to identify loci associated with resistance/susceptibility to infection with the nematode Angiostrongylus costaricensis. Parental strain A/J shows a high survival rate (90.5%) 5 weeks post-infection whereas parental strain SM/J shows a low survival rate (33.3%). Infected SM/J animals exhibit severe anemia and reduced body weight. A locus named Acsns1, Angiostongylus costaricensis nematode susceptibility 1, mapped to distal mouse Chromosome 19 from 47 cM (D19Mit91) to 53 cM (D19Mit34) with a LOD=4.3. Homozygosity for A/J-derived alleles appears to increase survival in infected F2 animals. A potential candidate gene is Csf2ra at 51 cM on mouse Chromosome 19. Epistatic interacting loci were detected at D13Mit248 and D17Mit21. The Chromosome 13 locus is named Acsns2 and the Chromosome 17 locus is named Acsns3. Homozygosity for SM/J-derived alleles at both Acsns2 and Acsns3 confers decreased host survival (P<0.001). J:7891322 SMXA (SM/J x A/J) recombinant inbred (RI) strains were analyzed for loci associated with susceptibility to infection with Angiostrongylus costaricensis. Parental strain A/J is resistant whereas parental strain SM/J is susceptible. 789 polymorphic loci were screened. Acsns2 mapped to 33 cM on mouse Chromosome 13 with P=0.0096 at D13Rik79 and D13Rik80. Acsns3 mapped to 18.4 cM on mouse Chromosome 17 with P=0.0088 between D17Mit28 and D17Rik76. Other yet unnamed loci associated with susceptibility to infection with Angiostrongylus costaricensis include D6Rik86 and D6Rik87 (P<0.0001), and D10Rik66 - D10Mit12 (P=0.0058). |
