Mapping and Phenotype information for this QTL, its variants and associated markers

J:76486

Linkage analysis was performed on 186 (C57BL/6J-Fcrg2btm1Rav x BALB/c-Fcrg2btm1Rav)F2 intercross animals to identify loci linked to autoimmune susceptibility. 81 microsatellite markers at an average resolution of 20 cM were screened. Parental strain C57BL/6J-Fcrg2btm1Rav develops spontaneous systemic lupus erythematosus (SLE) whereas BALB/c-Fcgr2btm1Rav does not. F1 hybrid animals do not develop SLE, similar to resistant parental strain BALB/c-Fcgr2btm1Rav. Autoimmune traits measured were antinuclear antibody (ANA) production, splenomegaly, and proteinuria. Autoimmune susceptibility loci mapped to mouse Chromosomes 9 (Asbb1), 12 (Asbb2), and 17 (Asbb3). Asbb1 is linked to splenomegaly (LOD=3.62) and spans 0 cM - 14 cM on mouse Chromosome 9. BALB/c-derived alleles confer recessively-inherited autoimmune susceptibility at Asbb1. Asbb2 is linked to ANA (LOD=3.87) and spans 2.5 cM - 8.5 cM on mouse Chromosome 12. C57BL/6J-derived alleles confer additively-inherited autoimmune susceptibility at Asbb2. Asbb3 is linked to ANA (LOD=7.58), proteinuria (LOD=3.62), and splenomegaly (LOD=2.68), and spans 5.5 cM - 23.5 cM on mouse Chromosome 17. Asbb3 gives peak linkage at D17Mit198 (16 cM) with the C57BL/6J-derived allele contributing recessive autoimmune susceptibility. Loci with suggestive linkage to ANA mapped to D1Mit60 and D7Mit117.
