Mapping and Phenotype information for this QTL, its variants and associated markersJ:26734The authors use F2 progeny of the B10.RIII x RIIIS/J cross to identify QTLs associated with susceptibility to chronic experimental autoimmune encephalomyelitis (EAE). RIIIS/J mice are resistant to induction of disease; B10.RIII mice develop a chronic disease. The Eae2 QTL was associated with the D15Nds2 locus on mouse Chromosome 15. J:96204Linkage analysis was performed on 402 animals from a B10.RIII-H2r/SgKljLund x RIIIS/J)F1 x B10.RIII-H2r/SgKljLund backcross to identify QTLs associated with experimental autoimmune encephalomyelitis (EAE) traits. Parental strain B10.RIII-H2r/SgKljLund is susceptible to developing EAE upon immunization with myelin basic protein (MBP) 89-101 peptide whereas parental strain RIIIS/J is resistant. 150 microsatellite markers were used for the genome scan. On mouse Chromosome 15, a locus including Eae2 (15.5 cM) showed linkage to EAE development (area under the curve-AUC) between markers D15Mit100 (21 cM) and D15Mit234 (34.2 cM). Animals homozygous for B10.RIII-H2r/SgKljLund-derived alleles exhibit the highest AUC. This inheritance pattern is different from what was previously observed with Eae2. Therefore, this locus is thought to be distinct from Eae2 and has been assigned the symbol Eae2b.On mouse Chromosome 15, Eae2 shows linkage to CD4+/CD8+ T cell ratios in female animals. This phenotype is linked to marker D15Mit150 (12.1 cMc) with an LRS of 13.3. |
