Candidate Genes

Search for an Lfp1 candidate gene on distal mouse Chromosome 19 resulted in the consideration of Chuk (45 cM), a helix-loop-helix protein expressed in the hypothalamus. It is thought Chuk may play a role in the regulation of the circadian rhythm. Wheel running activity of heterozygous Chuk mutant mice was compared to wild type. No differences were found in the length of running activity time, angle of entrainment, or amount of running in constant darkness. Therefore, Chuk is not likely a viable candidategene for Lfp1.

Mapping and Phenotype information for this QTL, its variants and associated markers

QTLs associated with free running period were provisionally mapped in 289 animals from a (CS x C57BL/6)F2 intercross and confirmed in 192 animals from a (CS x MSM)F2 intercross. Parental strain CS exhibits unstable free running patterns and spontaneous rhythm splitting whereas parental strains C57BL/6 and MSM exhibit regular running patterns.

QTLs for long free running period were identified on mouse Chromosome 19 in the

(CS x MSM)F2 intercross:

Lfp1 maps from 50 cM - tel with a LOD = 4.61 at D19Mit33;

Lfp2 maps from 26 cM - 47 cM with a LOD = 4.26 at D19Mit5;

and Lfp3 maps from 0 cM - 26 cM with a suggestive LOD = 3.85 at D19Mit32.

CS-derived alleles appear to increase the length of the free running period at Lfp1, Lfp2, and Lfp3 in a recessively inherited fashion. No obvious candidate genes were identified at these loci.

A suggestive locus mapped to mouse Chromosome 12 from 25 cM - 45 cM with LOD = 3.24 at D12Mit4 in the (CS x C57BL/6)F2 intercross but did not reach statistical significance withthe (CS x MSM)F2 intercross.
