Notes |
Mapping and Phenotype information for this QTL, its variants and associated markersJ:7427035 C57BL/6J x DBA/2J (BXD) recombinant inbred (RI) strains were used to map loci associated with peripheral blood lymphocyte proportions. Parental strain C57BL/6J exhibits a higher concentration of circulating white blood cells compared to DBA/2J. Two loci linked to peripheral blood B lymphocyte proportion, Pbbcp1 and Pbccp2, mapped to mouse Chromosome 1. Pbbcp1 spans 62 cM - 64 cM and gives strongest linkage at 63.1 cM with LOD=4.1 at D1Mit188. Pbbcp2 gives strongest linkage at 48.4 cM with LOD=3.7 at D1Ncvs45. A locus linked to peripheral blood CD8 T lymphocyte proportion, Pbctlp1, mapped to mouse Chromosome 19 from 9 cM - 24 cM with LOD=3.8 at D19Mit28 (12 cM). A (C57BL/6J x SJL)F2 intercross population was used to confirm the QTLs mapped in the BXD RI strains. Parental C57BL/6J exhibits a significantly higher concentration of circulating white blood cells compared to SJL. Pbbcp1 was detected in the F2 cross, but not Pbbcp2 or Pbctlp1. Authors speculate C57BL/6J and SJL may not be polymorphic at Pbbcp2 and Pbctlp1. In the F2 cross, Pbbcp1 spanned 59.8 cMp - 70 cM and showed strongest linkage at Gli2 (63 cM) with LOD=5.6. Possible candidate genes for Pbbcp1 are Gli2, En1, and Bcl2.Three markers adjacent to the Pbbcp1 locus, D1Mit387 (62 cM), Gli2 (63 cM), and D1Mit419 (63.1 cM) all showed an association with peripheral blood ( PB) lymphocytes that express B220, |
