Mapping and Phenotype information for this QTL, its variants and associated markers

Loci associated with thymocyte proliferative response were mapped in 14 C57BL/6J x DBA/2J (BXD) recombinant inbred (RI) strains. 22 month old animals were treated with concanavalin A (Con A) to induce thymocyte proliferation. Parental strain DBA/2J exhibits a high thymocyte proliferative response when induced with Con A whereas parental strain C57BL/6J exhibits a low proliferative response. A BXD mapping panel of 620 nonredundant markers were used to screen the animals. QTLs significantly linked to thymocyte proliferation Thypr1, Thypr2, and Thypr3 were mapped to mouse Chromosomes 11, 1, and 3, respectively. Thypr1 spans a range of 5 cM - 21 cM on mouse Chromosome 11 and reaches a maximum LRS=24 at D11Mit51 (18 cM). Il12b at 19 cM on chromosome 11 has been identified as a potential candidate gene for Thypr1. Thypr2 spans a range of 26 cM - 34 cM on mouse Chromosome 1 and reaches a maximum LRS=33 at D1Mit214 (32.1 cM). Potential candidate genes for Thypr2 include Ctla4 (30.1 cM), Cd28 (30.1 cM), and Icos (32 cM). Thypr3 reaches a maximum LRS=35 at D3Mit304 (5.6 cM) when Thypr1 and Thypr2 are held constant. A possible candidate gene for Thypr3 is Il7 at 6.6 cM on mouse Chromosome 3. All loci are highly significant when analyzed by composite interval mapping using an assumed additive mode of inheritance.
