Mapping and Phenotype information for this QTL, its variants and associated markers

QTL analysis was performed on 340 (KK.Cg-Ay/TaJcl x PWK/RpRbrc)F2 animals to identify loci associated with static glucokinesis and/or dynamic glucokinesis. Parental strain KK.CC-Ay/TaJcl exhibits diabetes whereas parental strain PWK/RpRbrc is non-diabetic.

A major locus mapped to 46.5 cM - 51.55 cM on mouse Chromosome 7 with LOD=26 between D7Nds18 and D7Mit130 in linkage to HbA1c concentrations (a marker of recent blood glucose levels.) Nidd1K also showed linkage to several dynamic glucokinesis values with LOD=6. Hbb, Ucp2,and Upc3 map near Nidd1k and may be potential candidate genes.

Suggestive loci were also identified in linkage to body weight (Bwt1k), dynamic glucokinesis (Nidd5K, Nidd6k), and static glucokinesis (Nidd7k) with LOD>2.8.

Bwt1kmapped to 31.4 cM - 42.5 cM on mouse Chromosome 4 between D4Mit17 and D4Mit26,

and Nidd5k mapped to 36.9 cM on mouse Chromosome 1 at D1Mit380.

Nidd6k mapped to 10 cM - 57 cM on mouse chromosome 11 between D11Mit170 and D11Mit67.

Interestingly, PWK-derived alleles appear to have a diabetogenic effect at Nidd6k.

Nidd7k mapped to 65 cM - 86 cM on mouse Chromosome 5 between D5Mit68 and D5Mit169.
