Mutation details: The entire human AGT genomic sequence containing a C to T nucleotide substitution at codon 235 was inserted into the Hprt locus via homologous recombination. (This polymorphism results in a threonine to methionine substitution and has been reported in association with hypertension in humans.) Northern blot analysis showed high transgene expression in liver and kidneys, and lower levels of transgene expression in brain, white and brown adipose tissue, diaphram, and aorta. In addition, immunohistochemical analysis of kidney and liver sections detected transgene expression in the proximal tubule cells of the renal cortex and in hepatocytes.
