310 female (C57BL/6 x BXD-27/Ty)F2 animals were analyzed for QTLs associated with sensitivity to TPA-induced skin tumors. Parental strain C57BL/6 is resistant to TPA-induced skin tumors whereas parental strain BXD-27/Ty is susceptible. The strongest linkage was detected at 87 cM on mouse Chromosome 2 near D2Mit286 and was named Psl2 (promotion susceptibility QTL 2). DBA/2-derived alleles confer dominant susceptibility to TPA-induced tumors at this locus. A locus with an additive effect, Psl3 mapped to 111 cM on mouse Chromosome 1 near D1Mit511. C57BL/6-derived alleles confer susceptibility to TPA-induced tumors at this locus. Lastly, Psl4 mapped to 47 cM on mouse Chromosome 19 near D19Mit38. C57BL/6-derived alleles confer dominant resistance to TPA-induced tumors at Psl4.
