304 (C57BL/6J x DBA/2J)F2 animals and 261 (C57BL/6J x DBA/2J)F1 x DBA/2J backcross animals were screened for 84 microsatellite markers at an average spacing of 15 cM to map QTLs affecting CD4:CD8 T cell ratios. Parental strain C57BL/6J exhibits low CD4:CD8 T cell ratios compared to parental strain DBA/2J.

Trmq1 (T cell ratio modifier QTL 1) mapped to 74 cM on mouse Chromosome 6 with a LOD score of 4.6 at D6Mit15 in the F2 cross. The flanking markers for Trmq1 are D6Mit198 (67 cM) and D6Mit15 (74 cM). DBA/2J-derived alleles confer increased lymph node CD4:CD8 levels with dominant inheritance. Candidate genes mapping to the QTL interval of Trmq1 are Klra, Klrb1c, and Klrb1.

Trmq2 mapped to 52 cM on mouse Chromosome 2 with a LOD score of 3.4 at D2Mit483 andD2Mit207 in the backcross population. The flanking markers for Trmq2 are D2Mit61 (34 cM) and D2Mit17 (69 cM). C57BL/6J-derived alleles confer increased lymph node CD4:CD8 levels with dominant inheritance. Candidate genes mapping to the QTL interval of Trmq2 are CD44, Rag1, Rag2, H3, Brm, Ir2, Il1a, Il1b, H42, H44, H45, and Ir9.
