Mapping and Phenotype information for this QTL, its variants and associated markers

J:78523

The Tg(Igh-Abl1)2Sco transgene was constructed on a (C57BL/6JWehi X SJL/JWehi)F1 background and was transferred onto a C57BL/6J background and a BALB/cAnBrdWehi background by sucessive backcrossing. Transgenic animals on the BALB/cAnBrdWehi background are more susceptible to plasmatycoma development than transgenic animals on a C57BL/6J background.

Transgenic C57BL/6J animals were crossed to BALB/c mice. Transgenic F2 animals were susequently generated. These animals were screened for 191 polymorphic markers at an average resolution of 10 cM to identify QTLs associated with susceptibility to plasmacytoma.

Plasmacytoma susceptibility 1 (Pcyts1) mapped to 69 cM on mouse Chromosome 2 with a peak LOD score of 6.1 at D2Mit17. BALB/c-derived alleles confer plasmacytoma susceptibility in male animals with additive inheritance. Possible interaction was detected between Pcyts5-Pcyts1.

Pcyts2 maps to 12 cM on mouse Chromosome 4 with a peak LOD score of 5.2 at D4Mit236. BALB/c-derived alleles confer additively inherited plasmacytoma susceptibility.

Pcyts3 maps to 49 cM on mouse Chromosome 11 with a peak LOD score of 5.5 at D11Mit39. BALB/c-derived alleles confer dominantly inherited plasmacytoma susceptibility at Pcyts3. Possible interaction was detected between Pcyts5-Pcyts3.

Pcyts4 and Pcyts5 mapped to 8 cM and 34 cM on mouse Chromosome 17, respectively. Pcyts4 gives a peak LOD score of 5.6 at D17Mit57 with BALB/c-derived alleles conferring additively inherited plasmacytoma susceptibility in female animals. Pcyts5 gives a peak LOD score of 6.6 at D17Mit87 with BALB/c-derived alleles conferring additively inherited plasmacytoma susceptibility in female animals. Possible interactions were detected between Pcyts5-Pcyts1 and Pcyts5-Pcyts3.

Pcyts6 maps to 25cM on mouse Chromosome 18 with a peak LOD score of 7.8 at D18Mit74. C57BL/6J-derived alleles confer additively inherited plasmacytoma susceptibility at Pcyts6.
