Mapping and Phenotype information for this QTL, its variants and associated markersJ:82708A replicate population of 112 female and 94 male (MRL/MpJ-Faslpr x C57BL/6J)F2 animals was screened for 138 polymorphic markers at an average spacing of 9.5 cM +/- 6.6 cM to examine and confirm QTLs associated with wound healing. Parental strain MRL/MpJ-Faslpr exhibits faster wound healing after introduction of an ear hole punch compared to C57BL/6J. Interestingly, MRL/MpJ female animals exhibit faster wound healing than MRL/MpJ males, but these effects are not observed in the MRL/MpJ-Faslpr substrain. Several previously identified wound healing QTLs were confirmed in this study, and 3 new loci were identified. Heal1 (Chr 8), Heal2, Heal3, and Heal7 (Ch 13), and Heal6 (Chr 7), are previously identified QTLs reaching statistical significance inthis study. 03.03.2015 Curators Note: Because Heal1, Heal2, Heal3, Heal7 and Heal6 were previously mapped using an (MRL/MpJ-Faslpr x C57BL/6NTac)F2 intercross (J:50111, 1998) and C57BL/6NTac is a substrain of the C57BL/6J mice used here, we consider this a separate map study and have named the QTL identified here as Heal14 (Chr8), Heal17, Heal18, Heal21 (Chr13) and Heal20 (Chr7) respectively.TABLE 1: Summary of (MRL xB6)F2QTL Heal14 mapped between 49 cM - 68 cM on mouse Chromosome 8. Peak linkage occured at D13Mit117 with LRS=15.2. The healing allele was inherited from the MRL parent in a dominant mode of inheritance. Males exhibit increased wound healing compared to females at this locus.QTL Heal18 mapped between 48 cM - 60 cM also on mouse Chromosome 13. Peak linkage occured at D13Mit144 with LRS=14.9. The healing allele was inherited from the MRL parent in an additive mode. Male mice also exhibited increased healing at this locus compared to female mice.QTL Heal20 mapped between 26 cM - 50 cM on mouse Chromosome 7. Peak linkage occured at D7Mit85, LRS=5.1. The healing allele was inherited from the MRL parent in an additive mode of inheritance and displayed no sexaul dimorphism.QTL Heal21 mapped between 30 cM - 35 cM on mouse Chromosome 13. Peak linkage occured at D13Mit245, LRS=21.9. The healing allele was inherited from the MRL parent in a dominant mode of inheritance with male mice exhibiting increased healing at this locus compared to female mice.Heal8 was previoulsy mapped to mouse Chromosome 13 using an (MRL/MpJ-Faslpr x C57BL/6NTac)F2 intercross (J:50111, 1998). This is a separate map study and we have named this QTL, mapping to mouse Chromosome 4, between 66 cM - 71 cM, Heal22. Peak linkage occured at D4Mit13, LRS=13.8. The MRL/MpJ derived alleles confer recessively inherited increase in wound with male mice exhibiting increased healing at this locus compared to female mice.Novel QTL Heal9 mapped to 30 cM - 44 cM on mouse Chromosome 18. Peak linkage occurred at D18Mit33 (44 cM) with LRS=5.7.Interestingly, the heterozygous state confers increased wound healing compared to homozygosity for either parental allele. Females exhibit increased wound healing compared to males at Heal9.Novel QTL Heal10mapped to 55 cM - 62 cM on mouse Chromosome 11. Peak linkage occurred at D11Mit213 (55 cM) with LRS=11. MRL/MpJ-derived alleles conferring additively inherited increase in wound healing. Males exhibit increased wound healing compared to females at Heal10. Novel QTL Heal11 mapped to 4 cM - 20 cM onmouse Chromosome 16. Peak linkage occurred at D16Mit122 with LRS=9.1. C57BL/6J-derived alleles confer recessively inherited increase in wound healing. Male exhibit increased wound healing compared to females at Heal11. |
