Mapping and Phenotype information for this QTL, its variants and associated markers

170 (BALB/cByJ x DBA/2J)F1 x BALB/cByJ backcross animals were genotyped for 199 polymorphic markers to identify QTLs associated with sucsceptibility to demyelinating disease caused by Theiler's murine encephalomyelitis virus. Parental strain DBA/2J is susceptible to Theiler's murine encephalomyelitis virus induced demyelinating disease whereas parental strain BALB/cByJ is resistant. Four new loci, designated Tmevd6, Tmevd7, Tmevd8, and Tmevd9, were identified in this study.

Tmevd6 maps to 19.5 cM on mouse Chromosome 1 near D1Mit170 and spans approximately 10 cM - 26 cM. This locus accounts for 9.5% of the phenotypic variation. Interestingly, the BALB/cByJ-derived allele confers increased disease severity in males while the DBA/2J-derived allele confers decreased disease severity in females. This QTL may contain two tightly linked genes conferring opposite effects in males and females.

Tmevd7 maps to 72 cM on mouse Chromosome 5 near D5Mit30 and spans approximately 70 cM - 81 cM. This locus accounts for 16.6% of the phenotypic variation. DBA/2J-derived alleles confer increased disease severity in male animals at Tmevd7.

Tmevd8 maps to 4.7 cM on mouse Chromosome 15 near D15Mit12 and spans approximately 4 cM - 15 cM. This locus accounts for 14.1% of the phenotypic variation. DBA/2J-derived alleles confer susceptibility in male animals at Tmevd8.

Tmevd9 maps to 32.8 cM on mouse Chromosome 1 near D1Mit76 and spans approximately 32 cM -43 cM. This locus accounts for 7.6% of the phenotypic variation. DBA/2J-derived alleles confer decreased disease severity in female animals at Tmevd9. Cd28 and Ctla4 are 2 possible candidate genes mapping near Tmevd9.

Suggestive QTLs were detected at 22 cM on mouse Chromosome 15 near D15Mit5 and at 53.5 cM on mouse Chromosome 16 near D16Mit50. Both of these loci affect disease severity in male animals. The chromosome 16 locus colocalizes with Eae11, a previously identified QTL associated with susceptibility to experimental allergic encephalomyelitis.
