Mutation details: A loxP flanked cassette containing neo and tk genes replaced a 257 bp fragment of intron 7, eliminating a portion of the internal promoter that drives expression of two short isoforms, TRdeltaalpha1 and TRdeltaalpha2. The floxed selection genes were excised prior to germline transmission. Whereas wild-type levels of transcript encoding the two longer isoforms, TRalpha1 and TRalpha2, were detected in homozygous mutant mice, the levels of transcript encoding the shorter isoforms were reduced.
