Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 470 animals from a (C57BL/6J x KJR/Ms)F2 cross to identify QTLs associated with capsaicin sensitivity. (Capsaicin is a compound found in hot chili peppers which causes a burning sensation.) Genome scan was performed using 124 markers at an average spacing of 20 cM. Parental strain C57BL/6J is more sensitive to capsaicin compared to parental strain KJR/Ms. (KJR/Ms consumes more capsaicin solution compared to C57BL/6J.) F1 hybrids resemble KJR/Ms and consume greater amounts of capsaicin solution compared to C57BL/6J.

Capsq1 (capsaicin sensitivity related QTL 1) maps between D2Mit120 (17 cM) and D2Mit126 (47.5 cM) on mouse Chromosome 2. Capsq1 is linked to capsaicin consumption with LOD=5.88 for 4 uM capsaicin solution, LOD=4.87 for 7 uM capsaicin solution, and LOD=5.93 for 10 uM capsaicin solution. KJR/Ms-derived alleles confer increased capsaicin consumption (decreased capsaicin sensitivity) at this locus.

Capsq2 maps between D7Mit191 (3.4 cM) and D7Mit69 (24.5 cM) on mouse Chromosome 7. Capsq2 is linked to 4 uM capsaicin consumption with LOD=4.79, and is also suggestively linked to 7 uM capsaicin consumption. KJR/Ms-derived alleles increased capsaicin consumption (decreased capsaicin sensitivity) at this locus.

Capsq3maps between D7Mit31 (44 cM) and D7Mit66 (57.5 cM) on mouse Chromosome 7. Capsq3 is linked to 10 uM capsaicin consumption with LOD=5.76, and is also suggestively linked to 1 uM and 7 uM capsaicin consumption. KJR/Ms-derived alleles increased capsaicin consumption (decreased capsaicin sensitivity) at this locus.

Capsq4 maps between D8Mit293 (21 cM) and D8Mit240 (43 cM) on mouse Chromosome 8. Capsq4 is linked to 10 uM capsaicin consumption with LOD=4.42, and is also suggestively linked to 7 uM capsaicin consumption. Interestingly, animals homozygous for KJR/Ms- and C57BL/6J-derived alleles at Capsq4 exhibit the same amount of capsaicin consumption while heterozygous animals exhibit decreased capsaicin consumption.
