Genome scan at a resolution of 15 cM was performed on 3 different F2 intercross populations to identify QTLs associated with basal locomotor activity. The F2 populations used were (C57BL/6J x DBA/2J)F2, (C57BL/6J x BALB/cJ)F2, and (C57BL/6J x LP/J)F2.

A QTL for basal locomotor activity was detected on distal mouse Chromosome 1 and named Bslm4. Bslm4 shows linkage to basal locomotor activity at D1Mit150 (100 cM) with LOD=6.5 in the (C57BL/6J x DBA/2J)F2 cross, LOD=4.7 in the (C57BL/6J x BALB/cJ)F2 cross, and LOD=3.9 in the (C57BL/6J x LP/J)F2. C57BL/6J-derived alleles confer increased basal locomotor activity at Bslm4.

Furthermore, Bslm4 was confirmed using gene expression, SNP, and congenic strain data. Candidate genes mapping the Bslm4 interval include Aldh9a1, Rgs5, Kcnj10, and Kcnj9. Expression data showed differential expression of Kcnj9 in the brain with C57BL/6J exhibiting lower expression compared to DBA/2J, BALB/cJ, and LP/J. When Kcnj9 expression was analyzed in a BXD RI mapping panel a LOD score > 12 was attained. Kcnj10 is closely linked to Kcnj9 but exhibits suggestive linkage. SNP data showed a high likelihood of a QTL located between 168 Mb - 178 Mb on mouse Chromosome 1 and centered at 173 Mb. Analysis of the B6.D2-Mtv7/Ty congenic strainshows that animals homozygous or heterozygous for DBA/2J-derived alleles in the Mtv7 interval (90 cM - 100 cM on chromosome 1) exhibit decreased locomotor activity compared to animals homozygous for C57BL/6J-derived alleles.
