Mutation details: The frt-flanked neo cassette was excised from Efnb2tm1Henk via FLP-mediated recombination, leaving the loxP flanked lacZ cassette in exon 5. The lacZ cassette disrupted codons 264 through 340 which make up the majority of the cytoplasmic domain. Biochemical analysis indicated that the expected EFNB2-beta-gal protein, in which the endogenous cytoplasmic domain is replaced with beta-gal, was produced. This mutation leaves forward signaling intact and ablates reverse signaling. The expression pattern of lacZ was observed to be to similar to that of the endogenous gene.
