Mutation details760bp from the coding region were replaced with lacZ and a neomycin resistance gene. The replaced sequence included the translation initiation codon and sequence coding for the first 5 transmembrane spanning domains. RT-PCR indicated that less than 10% of normal transcript levels were present and this was most likely the consequence of read-through from the neomycin resistance gene rather than wild-type transcript.
