A genome scan was performed on (C57BL/6 x C3H/HeJCr)F2 intercross animals to confirm previously identified QTLs affecting proteoglycan-induced arthritis (Pgia1-12) and identify new QTLs. C3H/HeJCr inbred strain was newly identified as an arthritis susceptible strain following proteoglycan challenge. Inbred strain C57BL/6 is arthritis resistant after proteoglycan challenge. Pgia5 (chromosome 9, LOD = 4.35 at 32 cM) and Pgia10 (chromosome 16, LOD = 3.90 at 30 cM) were confirmed as QTLs affecting proteoglycan-induced arthritis. Pgia5 shows linkage to Bbaa9, a locus associated with IgG production. Novel QTLs Pgia13 (Chromosome 4, LOD = 4.40 at 2 cM), Pgia14 (Chromosome 12, LOD = 4.55 at 54 cM), Pgia15 (Chromosome 13, LOD = 3.95 at 25 cM), and Pgia17 (Chromosome 17, LOD = 9.15 at 12 cM) were identified. A suggestive QTL, Pgia16 mapped to Chromosome 5 at 36 cM (LOD = 2.95). Pgia5 and Pgia13 are linked to disease onset and Pgia10, Pgia14, and Pgia15 are linked to disease severity. All PGIA QTLs are recessive with the exception of Pgia17, which shows additive inheritance. Various PGIA QTLs colocalization with QTLs mapped for other phenotypes: Pgia13 showed linkage to Lmb1 in the mouse model for lupus, Pgia14 showed linkage to Pia3 for pristine-induced arthritis in rats, Pgia15 showed linkage to Cia11 (Mcia3) for collagen-induced arthritis, and Pgia16 showed linkage to Bbaa2, a locus associated with ankle swelling in the Lyme disease model.
