Linkage analysis using 73 polymorphic markers was performed on 83 (B10.D2 x BALB/cJ)F1 x BALB/cJ backcross animals to identify QTLs associated with T helper (Th) cell response. (*Authors did not indicate which specific strain of B10.D2 was used in this study.) Parental strain C57BL/10 develops a predominantly Th1 response whereas parental strain BALB/c develops a predominantly Th2 response. (B10.D2 x BALB/cJ)F1 hybrid animals develop a predominantly Th1 response similar to the C57BL/10 parent.

A locus associated with Th cell response, Thcr, mapped between D18Mit149 (24 cM) and D18Mit207 (41 cM) on mouse Chromosome 18. This locus is associated with increased Ifng levels. Thcr was confirmed in congenic animals carrying a region spanning 11 cM - 41 cM from B10.D2 introgressed onto the genetic background of BALB/cJ. These congenics exhibit decreased IL-4 production and increased Ifng production. Fine-mapping localized Thcr between 54.1 cM and 57.8 cM.

Suggestive linkage to Ifng levels mapped to mouse Chromosome 12 between 20 cM - 50 cM (D12Mit285 and D12Mit277, respectively; LOD=1.6). Suggestive linkage to IL-5 levels mapped to mouse Chromosome 14 between 19.5 cM and 44.3 cM (D14Mit142 and D14Mit195, respectively; LOD=1.2) and to mouse Chromosome 5 between 59 cM and 78 cM (D5Mit314 and D5Mit31, respectively; LOD=1.2).
