Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on a population of (C57BL/6J x DBA/2J)F2 animals at a resolution of 13 cM to identify QTLs associated with aortic lesion size and lipid traits. Animals were placed on an atherogenic diet for 16 weeks and examined. Parental strain C57BL/6J exhibits larger aortic lesions compared to parental strain DBA/2J.

Significant linkage was detected on mouse Chromosome 10 near D10Mit42 (LOD=4.5). *Note: Authors give the position of D10Mit42 at 24 cM whereas in MGI this marker is assigned to 44 cM.* This locus is named Aorls1 (aortic lesion size 1) and explains 11% of the phenotypic variance. DBA/2J-derived alleles confer increased aortic lesion size at Aorls1. Animals homozygous for C57BL/6J-derived alleles at Aorls1 exhibit 1/3 the lesion size of DBA/2J homozygous animals. Aorls1 maps near Ath11, a previously identified QTL associated with lesion size.

Aorls2 mapped to 24.5 cM on mouse Chromosome 7 (LOD=3.67 at D7Mit193). C57BL/6J-derived alleles confer increased aortic lesion size at Aorls2. This locus explains 10% of the phenotypic variance.

A locus suggestively associated with atherosclerotic lesion size mapped to mouse Chromosome 8 near D8Mit41 (LOD=3.39) with C57BL/6J-derived alleles conferring increased lesion size.

Hdl5 mapped to 33 cM (D3Mit241) on mouse Chromosome 3 in linkage with total cholesterol (LOD=6.96), HDL cholesterol (LOD=5.33), and LDL cholesterol (LOD=4.43) levels on a CHOW diet. C57BL/6J-derived alleles confer increased cholesterol levels at Hdl5.

Hdl6 mapped to 65 cM (D6Mit25) on mouse Chromosome 6 in linkage to total cholesterol (LOD=4.3) and HDL cholesterol (LOD=6.98) levels on a CHOW diet. DBA/2J-derived alleles confer increased cholesterol levels at Hdl6.

Ffal1 mapped to 54 cM (D17Mit132) on mouse Chromosome 17 in linkage to free fatty acid levels (LOD=4.4) on a CHOW diet. C57BL/6J-derived alleles confer increased free fatty acids at Ffal1. This locus is also suggestively linked to total cholesterol on a CHOW diet (LOD=2.97).

A locus suggestively linked to HDL levels on a CHOW diet mapped to mouse Chromosome 18 near D18Mit50 (LOD=3.02). A locus suggestively linked to HDL levels on an atherogenic diet mapped to mouse Chromosome 2 near D2Mit50 (LOD=3.09). Loci suggestively linked to LDL levels on a CHOWdiet mapped to mouse Chromosome 2 near D2Mit63 (LOD=3.27) and to mouse Chromosome 11 near D11Mit199 (LOD=2.82). Loci suggestively linked to triglyceride levels on a CHOW diet mapped to mouse Chromosome 2 near D2Mit495 (LOD=3.03), mouse Chromosome 5 near D5Mit1 (LOD=4.26), and to mouse Chromosome 11 near D11Mit54 (LOD=3.6).
