22 LSxSS recombinant inbred (RI) strains were analyzed for loci associated with altered body temperature in response to dietary restriction. Decreased body temperature is a hallmark of dietary restriction, which is also associated with life extension. Dietary restricted animals were fed 60% the amount of food as ad libitum control animals. 72 markers at an average spacing of 20 cM were used in the genome scan.

A significant locus named Tbdr1 (body temperature response to dietary restriction, QTL 1) mapped to 29 cM on mouse Chromosome 9 with peak linkage at D9Mit4 (LOD=3.1). SS-derived alleles confer decreased body temperature at Tbdr1. This locus explains approximately 49% of the genetic variance. The 95% confidence interval of Tbdr1 spans from D9Mit228(26 cM) to D9Mit6 (31 cM). Nine candidate genes in this region show altered expressed in dietary restricted animals. These genes are H2afx, Hyou1 (Cab140), Apoa1, Apoa4, Zfp145 (Plzf), Drd2, Cyp19a1, Dnaja4, and Csk.

A suggestive locus mapped to 24 cM on mouse Chromosome 17 between D17Mit49 (23.2 cM) and D17Mit10 (24.5 cM) with LOD=1.5. This locus explains approximately 17% of the genetic variance with SS-derived alleles conferring decreased body temperature.
