Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 179 (B6.C-H25c/(HW65)ByJ x BALB/cJ)F2 intercross animals to map QTLs associated with plasma cholesterol levels. 110 polymorphic markers at an average spacing of 12 cM were typed. Parental strain B6.C-H25c/(HW65)ByJ exhibits increased HDL cholesterol, total cholesterol, and non-HDL cholesterol compared to parental strain BALB/cJ. B6.C-H25c/(HW65)ByJ also exhibits elevated apoA2 and apoE, but decreased apoA1 and apoC3 levels, compared to BALB/cJ.

A female-specific locus named Lipq1 mapped to74 cM on mouse Chromosome 6 near D6Mit15. Lipq1 is associated with HDL cholesterol level (LOD=5.0), total cholesterol level (LOD=6.3), and non-HDL cholesterol level (LOD=3.7). C57BL/6By-derived alleles confer increased HDL cholesterol, total cholesterol, and non-HDL cholesterol with additive inheritance at Lipq1. This locus was confirmed in a congenic strain (B6.NODc6) carrying NOD-derived donor DNA covering the Lipq1 locus on a C57BL/6 background. Female congenic animals exhibitelevated HDL cholesterol and decreased non-HDL cholesterol compared to C57BL/6 controls. The QTL interval of Lipq1 extends from 64 cM to the telomere (approximately 10 cM in distance).

A male-specific locus named Lipq2 mapped to 40 cM on mouse Chromosome 13 near D13Mit11. Lipq2 is associated with HDL cholesterol level (LOD=4.8) and total cholesterol level (5.5). BALB/cJ-derived alleles confer increased HDL and total cholesterol with recessive or additive inheritance at Lipq2. The QTL interval of Lipq2 extends approximately 13 cM. Two positional candidate genes mapping within the Lipq2 interval are Hmgcr and Hmgcs1.

Lipq3 mapped to 49.6 cM on mouse Chromosome 15 near D15Mit159. This locus is associated with HDL cholesterol level (LOD=5.2) and total cholesterol level (LOD=5.4). C57BL/6By-derived alleles confer increased HDL cholesterol and total cholesterol levels with dominant inheritance at Lipq3. Three positional candidate genes map within the Lipq3 interval: Ppara, Srebf2, and Dgat1.
