Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 380 animals from a (C57BL/6J x M. m. castaneus)F1 x M. m. castaneus backcross to identify QTLs associated with postnatal growth. A total of 13 novel QTLs were mapped. Pbwg1-Pbwg7 affect the entire growth period whereas Pbw8 and Pbw9 affect the mid- and early growth phases. M. m. castaneus-derived alleles confer reduced growth at Pbwg1-Pbwg7. Pbwg10-Pbwg13 were detected in epistatic interactions involving other Pbwg QTLs.

Pbwg1 (postnatal body weight growth 1) mapped to35 cM on mouse Chromsome 2 near D2Mit324 (LOD=8.9). The 95% confidence interval of Pbwg1 spans 26 cM - 44 cM. Pbwg1 appears to have a stronger effect in females compared to males. This locus explains 10% of the phenotypic variance. Gcg is a candidate genemapping near Pbwg1. Pbwg11 maps to 87 cM (D2Mit48) on mouse Chromosome 2 and interacts epistatically with Pbwg4 (P=1.8 x 10-5).

A male-specific QTL named Pbwg2 mapped to 62 cM on mouse Chromosome 4 near D4Mit32 (LOD=2.9). The 95% confidence interval of Pbwg2 spans 39 cM - 86 cM. This locus explains 3.9% of the phenotypic variance.

Pbwg3 maps to 72 cM on mouse Chromosome 7 with D7Mit259 (LOD=3.0). The 95% confidence interval of Pbwg3 spans 40 cM - 104 cM. This locus explains 3.0% of the phenotypic variance. Candidate genes mapping near Pbwg3 are Tub, Ucp2, and Ucp3.

A female-specific QTL named Pbwg4 maps to 71 cM on mouse Chromosome 9 with D9Mit18 (LOD=3.7). The 95% confidence interval of Pbwg4 spans 46 cM - 96 cM. This locus explains 7.5% of the phenotypic variance.

Pbwg5 maps to 68 cM on mouse Chromosome 10 near D10Mit145 (LOD=3.4). The 95% confidence interval of Pbwg5 spans 42 cM - 94 cM. This locus explains 3.7% of the phenotypic variance. Igf1 is a candidate genes mapping near Pbwg5.

Pbwg9 mapsto 14 cM on mouse Chromosome 10 near D10Mit16 (LOD=3.8). The 95% confidence interval of Pbwg9 spans 0 cM - 36 cM. Pbw9 appears to have a stronger effect in females compared to males. This locus explains 4.5% of the phenotypic variance.

Pbwg6 maps to 53cMonmouse Chromosome 13 near D13Mit195 (LOD=4.9). The 95% confidence interval of Pbwg6 spans 34 cM - 72 cM. Pbwg6 appears to have a stronger effect in males compared to females. This locus explains 5.2% of the phenotypic variance.

A male-specific locus named Pbwg7 maps to 19 cM on mouse Chromosome X with marker DXMit75 (LOD=3.1). The 95% confidence interval of Pbwg7 spans 0 cM - 48 cM. This locus explains 6.9% of the phenotypic variance.

Pbwg10 maps to 2.8 cM (DXMit124) on mouse Chromosome X and interacts epistatically with Pbwg8 (P=2.6 x 10-6). The effect of the Pbwg10-Pbwg8 interaction is male-specific.

A male-specific locus named Pbwg8 maps to 32 cM on mouse Chromosome 6 near D6Mit29 (LOD=4.). The 95% confidence interval of Pbwg8 spans 14 cM - 50cM. This locus explains 12.1% of the phenotypic variance. Ghrhr is a candidate genes mapping near Pbwg8.

Pbwg12 maps to 34 cM (D12Mit4) on mouse Chromosome 12 and interacts epistatically with Pbwg1 (P=9.1 x 10-6). The effect of the Pbwg12-Pbwg1 interaction is male-specific.

Pbwg13 maps to 81 cM (D5Mit51) on mouse Chromosome 5 and interacts epistatically with Pbwg9 (P=2.5 x 10-6). The effect of the Pbwg12-Pbwg9 interaction is female-specific.

Linkage analysis was performed on 387 (C57BL/6J x M. m. castaneus)F1 x M. m. castaneus backcross animals to identify QTL associated with postnatal body weight and growth traits. 90 polymorphic markers were used for the genome scan and average intermarker distance varied between 19.5 cM - 20.2 cM. Animals were weighed weekly between 3 - 10 weeks of age. Parental strain M. m. castaneus exhibits 60% smaller body size compared to parental strain C57BL/6J.

Eight previously mapped growth QTL (Pbwg1-Pbwg7, Pbwg9) were detected and re-confirmed in this study. Novel QTL Pbwg14-Pbwg22 were mapped in this experiment. Several of these loci participate in epistatic interactions. M. m. castaneus-derived alleles confer decreased body weight and/or weight gain at allloci except Pbwg9 and Pbwg19 where M. m. castaneus-derived alleles confer increased body weight and/or weight gain.

Pbwg1 mapped to 33 cM on mouse Chromosome 2 with marker D2Mit324 (LOD=3.1-10.9) and is associated with body weight at 4-10 weeks of age, weight gain from 3-6 weeks of age, and weight gain from 6-10 weeks of age. The minimal 95% confidence interval of Pbwg1 spans 17 cM.

Pbwg2 mapped to 62 cM on mouse Chromosome 4 near D4Mit32 (LOD=3.1) and is associated with body weight at 9 weeks of age. The 95%confidence interval of Pbwg2 spans 46 cM.

Female-specific QTL Pbwg14 mapped to 1 cM on mouse Chromosome 5 with marker D5Mit345 (LOD=2.8) and is associated with body weight at 8 weeks of age. The 95% confidence interval of Pbwg14 spans 76 cM.

Pbwg3 mapped to 72 cM on mouse Chromosome 7 with marker D7Mit259 (LOD=2.6-3.6) and is associated with body weight at 5-9 weeks of age. The minimal 95% confidence interval of Pbwg3 spans 56 cM.

Pbwg15 and Pbwg4 mapped to mouse Chromosome 9 at 43 cM and 69 cM, respectively. Pbwg15 peaks near D9Mit340 (LOD=2.6-2.7) and is associated with body weight at 3-4 weeks of age. Pbwg4 peaks near D9Mit18 (LOD=3.5) and is associated with body weight at 5 weeks of age. The minimal 95% confidence intervals for Pbwg4 and Pbwg15 span 62cM.

Pbwg5 mapped to 68 cM on mouse Chromosome 10 near D10Mit145 (LOD=2.6) and is associated with body weight at 7 weeks of age. The 95% confidence interval of Pbwg5 spans 76 cM. Pbwg9 and Pbwg16 mapped to 14 cM (LOD=3.5 at D10Mit16) and 45cM (LOD=4.6 at D10Mit20), repectively. Pbwg9 is associated with weight gain from 3-6 weeks of age and Pbwg16 is associated with body weight at 3 weeks of age. Unlike the other loci, M. m. castaneus-derived alleles at Pbwg9 confer increased weight gain.

Pbwg6 and Pbwg17 mapped to 53 cM (LOD=4.2-5.1 at D13Mit195) and 46 cM (LOD=4.2-4.9 at D13Mit195) on mouse Chromosome 13, respectively. Pbwg17 is associated with body weight at 6-10 weeks of age and Pbwg6 is associated with body weight at 5-10 weeks of age.The minimal95% confidence intervals of Pbwg17 and Pbwg6 spans 35 cM and 40 cM, respectively.

Female-specific QTL Pbwg18 mapped to 30 cM on mouse Chromosome 14 with marker D14Mit159 (LOD=4.3). The 95% confidence interval of Pbwg18 spans 42 cM and this locus is associated with weight gain from 6 to 10 weeks of age.

Male-specific QTL Pbwg19 mapped to 2 cM on mouse Chromosome 16 with marker D16Mit32 (LOD=3). The 95% confidence interval of Pbwg19 spans 69 cM and this locus is associated with body weight at5 weeks of age. Unlike the other QTL, M. m. castaneus-derived alleles at Pbwg19 confer increased body weight in males.

Pbwg20 mapped to 0 cM on mouse Chromosome 19 with marker D19Mit29 (LOD=3.1-6.3) and is associated with body weight at 3-4 weeks of age. The minimal 95% confidence intervalofPbwg20 spans 25 cM.

Male-specific QTL Pbwg7 and Pbwg21 mapped to mouse Chromosome X at 17 cM and 27 cM, repectively. Pbwg7 is associated with body weight at 5-8 weeks of age and weight gain between 3-6 weeks of age(LOD=3.4-3.8 at DXMit75). The minimum 95% confidence interval of Pbwg7 spans 44 cM. Pbwg21 is associated with body weight at 3 weeks of age and weight gain between 3-6 weeks of age (LOD=2.6-3.6 at DXMit75).

Female-specific QTL Pbwg22 mapped to 35 cM onmouse Chromosome X near DXMit19 (LOD=2.9-4.3). The minimal 95% confidence interval of Pbwg22 spans 39 cM and this locus is associated with body weight at 9-10 weeks of age.
