Mapping and Phenotype information for this QTL, its variants and associated markersJ:8829985 (C3H/HeJ x C57BL/6J)F2 intercross animals were genotyped at 87 polymorphic loci to identify QTLs associated with susceptibility to dental caries. Parental strain C3H/HeJ is resistant to dental caries after inoculation with Streptococcus mutans serotype C whereas C57BL/6J is susceptible. Highly significant linkage was discovered between 50.3 cM (D8Mit211) and 74.3 cM (D8Mit280) on mouse Chromosome 8 (LRS>26.6). This locus is named Dntcs1 (dental caries susceptibility 1). C3H/HeJ-derived alleles confersusceptibility to dental caries at this locus. Potential candidate genes mapping near Dntcs1 include Kcnn1, Calr, Cacna1a, Nfatc3, and Efcbp2 (Necab2).Significant linkage was detected between 28 cM (D2Mit237) and 52.5 cM (D2Mit101) on mouse Chromosome 2(LRS>18.5). This locus is named Dntcs2. C57BL/6J-derived alleles confer susceptibility to dental caries at this locus. A potential candidate gene mapping near Dntcs2 is Cacnb2.Suggestive linkages mapped to 32.8 cM (D1Mit21) on mouse Chromosome 1 and 57.5 cM (D7Mit66) on mouse Chromosome 7. C57BL/6J-derived alleles confer susceptibility to dental caries at D1Mit21 while C3H/HeJ-derived alleles confer susceptibility to dental caries at D7Mit66. |
