Linkage analysis was performed on 24 BXD (B=C57BL/6J, D=DBA/2J) recombinant inbred (RI) strains to identify QTLs associated with thermoregulation after ethanol administration. Parental strain C57BL/6J displays greater decrease in core body temperature following ethanol administration compared to DBA/2J. Animals were administered ethanol via intraperitoneal injection at three different doses: 1.5 g/kg, 2.5 g/kg, and 3.5 g/kb. Four loci reaching statistical significance were identified: Ethm1 (ethanol induced thermoregulation 1) on chromosome 2, Ethm2 on chromosome 8, Ethm3 on chromosome 7, and Ethm4 on chromosome 11.

Ethm1 mapped to 69 cM on mouse Chromosome 2 near D2Byu3 and D2Ncvs56. Ethm1 showed the strongest association with decreased body temperature at 3.5 g/kg ethanol (P=0.0002). DBA/2J-derived alleles confer greater ethanol-induced drop in body temperature at this locus. Potential candidate genes mapping near Ethm1 include Adra2b (71 cM), Il1 (73 cM), Il1a (73 cM), Il1b (73 cM), and Avp (73.2 cM).

Ethm2 mapped to 33 cM on mouse Chromosome 8 near D8Rp1. Ethm2 shows the strongest association with decreased body temperature at 1.5 g/kg ethanol (P=0.0010). C57BL/6J-derived alleles confer greater ethanol-induced drop in body temperature at this locus. Potential candidate genes mapping near Ethm2 include Clcn3 (32.2 cM), Jund1 (33 cM), and Npy1r (33 cM).

Ethm3 mapped to 53 cM on mouse Chromosome 7 near D7Mit7. Ethm3 is associated with decreased body temperature at 2.5 g/kg ethanol (P=0.0001). C57BL/6J-derived alleles confer greater ethanol-induced drop in body temperature. Potential candidate genes mapping near Ethm3 include Scnn1b (56 cM) and Scnn1g (56 cM).

Ethm4 mapped to 59 cM on mouse Chromosome 11 near D11Mit14 and Psmd3 (Tstap91a). Ethm4 is associated with disruption of regulation of body temperature at 1.5 g/kg ethanol (P=0.0006). C57BL/6J-derived alleles confer increased disruption of regulation at this locus. Potential candidate genes mapping near Ethm4 include Scn4a (64 cM) and Cacnb1 (58 cM). Ethm4 is located close to a previously identified QTL named Estq3 (estradiol regulated response QTL 3) at 57 cM.
