Homozygosity for B10.O20-derived alleles at this locus is associated with the absence of nuclear cytoplasmic invaginations (NCIs) in lung tumors.
Mapping and Phenotype information for this QTL, its variants and associated markersJ:68464Linkage analysis was performed on F2 crosses derived between 3 different OcB/Dem recombinant congenic strains (OcB-4/Dem, OcB-6/Dem, OcB-9/Dem) and O20/A to identify QTLs associated with susceptibility to lung tumors. (The OcB/Dem RC strains are derived from progenitor strains O20/A and B10.O20.) Parental strain O20/A is susceptible to lung tumors whereas parental strain B10.O20 is resistant. F2 animals were treated with ENU in utero and lung tumors were assessed for nuclear cytoplasmic invaginations (NCI) and lymphocytic infiltration (LI). Linkage to NCI was observed in the (OcB-6/Dem x O20/A)F2 cross at the Kras locus (P=0.0456) at 71 cM on mouse Chromosome 6. This locus is named Ncilt (nuclear cytoplasmic invaginations in lung tumors.) Inheritance of 2 B10.O20-derived alleles at Kras is strongly correlated with the presence of NCI in lung tumors whereas inheritance of 2 O20/A-derived alleles is strongly correlated with the absence of NCI in lung tumors. In this cross, the presence of NCIs is seen insmaller tumors whereas the presence of LI is seen in larger tumors. |
