Linkage analysis was performed on 192 (C57BL/6J x DBA/2J)F2 intercross animals to identify QTLs associated with granulocyte colony stimulating factor (G-CSF) induced splenomegaly. Parental strain DBA/2J exhibits a 2-fold increased in spleen size when administered G-CSF compared to parental strain C57BL/6J. 100 polymorphic markers at an average spacing of 10 cM was used for linkage analysis.

Significant association to G-CSF induced splenomegaly mapped to and interval on mouse Chromosome 1 between D1Mit181 (42 cM) and D1Mit265 (74.3 cM) with a peak LOD score of 7.9. C57BL/6J-derived alleles confer increased splenomegaly at this locus, which is named Gcsfis (G-CSF induced splenomegaly).

Gcsfis was confirmed using congenic strain B6.SJL-Ptprca Pep3b/BoyJ. This congenic strain carries SJL-derived DNA from D1Mit308 (62.1 cM) - D1Mit288 (71.5 cM) -which includes the Gscfis region- on a C57BL/6J genetic background. G-CSF treated C57BL/6J animals exhibit increased splenomegaly compared to G-CSF treated congenic animals, thus confirming the presence of a C57BL/6J-derived splenomegaly-promoting locus at Gcsfis.
