Mutation details: The allele was created with a targeting vector replacing and disrupting exons 1-5, which encode the catlytic domain, with a gene encoding LacZ fused to a nuclear localizing signal. The PGK-gpt/neomycin-resistance gene cassette was inserted in the reverse direction, downstrem of lacZ. Northern blot analysis of homozygous mutant fibroblasts show no detectable mRNA and absence of protein was demonstrated in Western blot of mutant embryos.
