In transgenic mice on a C57BL/6 background, expression of the transgene is restricted to the T cell lineage (no B-cell expression). Thymocytes, peripheral T-cells and activated T cells from these mice withstand prolonged culture in the absence of growth factors. Cells are resistant to killing by gamma-radiation, glucocorticoids, ionomycin, PMA, and sodium azide, but not complement, cytotoxic T cells or Fas ligand. Hemizygotes have a normal total T-cell count and thymic involution rate but show an enhanced response to immunization. This transgenic line displays no detectable autoimmunity. These mice serve as a robust source for the production of T-cell lines or hybridomas. |
