Mutation details: A functionally rearranged Tcra gene was used in the construction of this transgene. A recombined Tcra gene with Valpha2.3 and JalphaTA39 gene segments was cloned from the B10.PL hybridoma "172.10". This hybridoma was specific for the 11 amino-terminal residues of myelin basic protein (MBP). Crossing this line with the Tg(TCRB)C14Jg line produces a T cell with a transgenic TCR that has high affinity for the first 11 amino acids of MBP. In these doubly transgenic mice, 50-95% of CD3+ cells expressed the transgenic Tcra gene.
