Mutation details: A functionally rearranged Tcrb gene was used in the construction of this transgene A recombined Tcrb gene with Vbeta8.2/Dbeta2/Jbeta2.6 gene segments was cloned from the B10.PL hybridoma "172.10". This hybridoma was specific for the 11 amino-terminal residues of myelin basic protein (MBP). Crossing this line with the Tg(TCRA)B1Jg line produces a T cell with a transgenic TCR that has affinity for the first 11 amino acids of MBP. In these doubly transgenic mice, almost all of the CD3+ cells express the transgenic Tcrb gene.
