Mapping and Phenotype information for this QTL, its variants and associated markersJ:44389A (C57BL/6J x C3H/HeJ)F2 intercross was used in a selective genotyping strategy to identify QTLs associated with susceptibility to ozone induced lung inflammation. Inflammation-prone C57BL/6J animals experience 5-fold greater infiltration of polymorphonuclear neutrophils than inflammation-resistant C3H/HeJ animals when exposed to ozone. (C57BL/6J x C3H/HeJ)F1 animals and (C57BL/6J x C3H/HeJ)F1 x C3H/HeJ backcross animals exhibit a resistant phenotype similar to the C3H/HeJ parental, while (C57BL/6J x C3H/HeJ)F2 animals and (C57BL/6J x C3H/HeJ)F1 x C57BL/6J backcross animals exhibit a broad phenotype distribution within the range of both parentals. Inf2, a QTL contributing to subacute ozone induced inflammation, mapped to an interval on chromosome 17 between D17Mit125 (20.21 cM) and D17Mit10 (24.5 cM). Potential candidate genes in the region of Inf2 include Tnf (19.06 cM), Mcpt6 (11.7 cM), Mcpt7 (10.4 cM), and Sod2 (7.6 cM). Inf2 maps near Bhr3 (14 cM), a bronchial hyperrsponsiveness QTL (De Sanctis, J:29231). A suggestive QTL for lung inflammation mapped to chromosome 11 (P<2.15 x 10-3) between D11Mit20 (20 cM) and D11Mit12 (71 cM). This locus is named Inf1. A potential candidate gene for Inf1 is Nos1 (45.6 cM). No interaction between Inf2 on chromosome 17 and Inf1 on chromosome 11 could be detected. |
