Mapping and Phenotype information for this QTL, its variants and associated markersJ:93219Linkage analysis was performed on 124 (CAST/Ei x LT/SvKau)F1 x LT/SvKau backcross animals to identify QTLs associated with premature ovulation and oocyte arrest. Parental strain LT/SvKau exhibits a high proportion of diploid primary oocyte ovulation and oocyte arrest in meiotic metaphase I. This phenotype shows incomplete penetrance and variable expressivity. 59 polymorphic markers were used in the initial genome scan. Linkage to the phenotype was detected on mouse Chromosomes 1, 9, and 17. An additional37 polymorphic markers were used to refine these loci. The strongest linkage mapped to 58.7 cM on mouse Chromosome 1 with LOD=5 at D1Mit306. This locus is named Popoa (premature ovulation and primary oocyte arrest). The QTL interval of Popoa spans approximately 50 cM (D1Mit382) - 73 cM (D1Mit498). LT/SvKau-derived alleles confer increased metaphase I oocyte arrest at Popoa. Several candidate genes (Mtap2, Kif22-ps, Kif21b, Ncl, Serpine2, Bcl2, Psmb6-rs1) map near Popoa but sequence analysis did not reveal any polymorphisms between CAST/Ei and LT/SvKau.Suggestive linkage to metaphase I arrest mapped to 48 cM on mouse Chromosome 9 with LOD=2.5 at D9Mit181. Several candidate genes map to this region: Map2k1, Omt2a, Omt2b, Bcl2a1a, Mtap4, and Cdc25a. Sequence analysis did not reveal any polymorphisms between CAST/Ei and LT/SvKau.Oriq1 (31 cM) is a previously identified ovulation rate QTL mapping near this locus.A locus on mouse Chromosome 17 was detected near D17Mit209 in the initial genome scan but didnot produce a significant LOD score after typing of additional markers. |
