Linkage analysis was performed on 518 female (MRL/MpJ x SJL/J)F2 animals to identify QTLs associated with serum alkaline phosphatase (ALP) activity. 137 polymorphic markers were used for the genome scan. Parental strain MRL/MpJ exhibits significantly higher (91% increase) serum ALP activity compared to parental strain SJL/J. (MRL/MpJ x SJL/J)F1 hybrid animals exhibit high serum ALP activity similar to the MRL/MpJ parent. Alkaline phosphatase is thought to be involved in bone formation and shows a positive correlation to total body mass index and periosteal circumference.

Linkage to serum alkaline phosphatase activity mapped to 73 cM on mouse Chromosome 1 near D2Mit304 (LOD=3.8). This locus is named Salpa1 (serum alkaline phosphatase activity 1). MRL/MpJ-derived alleles confer increased serum ALP activity at Salpa1. Significant interaction was detected between Salpa1 and a locus at D12Mit136 (13 cM). Animals homozygous for SJL/J-derived alleles at Salpa1 and homozygous for MRL/MpJ-derived alleles at D12Mit135 exhibit increased serum ALP activity. The reverse is also true- animals homozygous for MRL/MpJ-derived alleles at Salpa1 and homozygous for SJL/J-derived alleles at D12Mit136 also exhibit increased serum ALP activity. Suggestive linkage to serum ALPactivity was detected at 86 cM near D2Mit285 (LOD=3.4).

Linkage to serum alkaline phosphatase activity mapped to 30 cM on mouse Chromosome 6 near D6Mit261 (LOD=12). This locus is named Salpa2. MRL/MpJ-derived alleles confer increased ALP activity at Salpa2 with dominant inheritance. Salpa2 explains 12.1% of the total variance. Significant interaction was detected between Salpa2 and a locus at D12Mit190 (28 cM). Animals homozygous for MRL/MpJ-derived alleles at Salpa2 and homozygous for MRL/MpJ- or SJL/J-derived alleles at D12Mit190 exhibit the highest levels of ALP activity. Salpa2 maps near a previously identified bone QTL named Fcsa1 at 37 cM, and a previously identified Igf1 serum level QTL named Igf1sl1 37 cM-51 cM.
